Advances in the construction and application of transdermal delivery systems

Int J Pharm. 2025 Dec 25:686:126306. doi: 10.1016/j.ijpharm.2025.126306. Epub 2025 Oct 30.

Abstract

In recent years, transdermal drug delivery has become a significant research focus for novel drug delivery methods. The primary challenge lies in overcoming the stratum corneum barrier, which impedes drug penetration. Enhancing drug transmittance and bioavailability and effectively breaching the skin barrier are critical challenges in developing transdermal methods. This paper reviews the principles and applications of various enhancement techniques, including penetration enhancers, microneedles, and advanced systems such as liposomes, electroporation, and iontophoresis. It compares the advantages and disadvantages of these systems and discusses future directions for transdermal delivery. The aim is to assist researchers in advancing transdermal delivery systems.

Keywords: Electroporation; Iontophoresis; Liposome; Microneedle; Penetration enhancers; Transdermal delivery system.
