Expression cloning exploiting PCR rescue of transfected genes

Abstract

Expression cloning in eukaryotic cells has allowed the isolation of many genes for which no protein or nucleic acid sequence information was available. In this paper we describe a model methodology for PCR-mediated rescue of transfected genes from eukaryotic cell lines after functional selection. This protocol should allow any readily transfectable cell line to serve as a host for functional selection of cDNA expression libraries.
