Network-based prediction of drug combinations.
Cheng F, et al. Nat Commun 2019. PMID 30867426 Free PMC article.
Here we propose a network-based methodology to identify clinically efficacious drug combinations for specific diseases. By quantifying the network-based relationship between drug targets and disease proteins in the human protein-protein interactome, we show the existence of six distinct classes of drug-drug-disease combinations. Relying on approved drug combinations for hypertension and cancer, we find that only one of the six classes correlates with therapeutic effects: if the targets of the drugs both hit disease module, but target separate neighborhoods. ...

Treatment of young adults with Philadelphia-negative acute lymphoblastic leukemia and lymphoblastic lymphoma: Hyper-CVAD vs. pediatric-inspired regimens
Siegel SE, et al. Am J Hematol 2018 - Review. PMID 30058716 Free article.
For young adults with acute lymphoblastic leukemia, pediatric-based regimens are likely to provide the following when compared to hyper-CVAD regimens: better disease control, less hospitalization time, diminished acute toxicities, decreased financial cost, more quality-adjusted life years, and fewer adverse late effects, such as infertility, myelodysplasia, and second malignant neoplasms. ...

Immune-related adverse events from combination immunotherapy in cancer patients: A comprehensive meta-analysis of randomized controlled trials.
Zhang B, et al. Int Immunopharmacol 2018 - Review. PMID 30130720
BACKGROUND: Although available evidence from clinical trials has shown that immune checkpoint inhibitors (ICIs) combination therapy can lead to a series of immune-related adverse events (irAEs), the overall risk of irAEs on combination therapy has yet not been systematically reported. ...The risk ratio for all-grade diarrhea and all-grade colitis for combination therapy was 1.95 (95% CI 1.54, 2.46; P < 0.00001) and 4.45 (95% CI 3.04, 6.51; P < 0.00001), respectively. ...

An overview of chemotherapy-related cognitive dysfunction, or 'chemobrain'
Moore HC. Oncology (Williston Park) 2014 - Review. PMID 25224480 Free article.
These effects appear to be most pronounced in the short-term, with improvement over time expected for most patients. Pharmacologic treatments do not have proven value in the management of chemotherapy-related cognitive dysfunction, but patients may benefit from cognitive rehabilitation. ...

Chemotherapy protocols and incidence of oral mucositis. An integrative review
Curra M, et al. Einstein (Sao Paulo) 2018 - Review. PMID 29694618 Portuguese, English. Free PMC article.
This review aimed to analyze the scientific production on severity of oral mucositis as an adverse effect of chemotherapy. To this end, we performed a search at PubMed databases combining the keywords "oral mucositis" and "chemotherapy protocol". ...To provide the best prevention and treatment for oral mucositis, it is essential to know the chemotherapy protocols used and their effects on the oral cavity....

[Validation of interaction databases in psychopharmacotherapy]
Hahn M and Roll SC. Nervenarzt 2018. PMID 28741067 German.
BACKGROUND: Drug-drug interaction databases are an important tool to increase drug safety in polypharmacy. There are several drug interaction databases available but it is unclear which one shows the best results and therefore increases safety for the user of the databases and the patients. ...DISCUSSION: The best results in this comparison were achieved by MediQ but this database also needs improvement with respect to the hit rate so that the users can rely on the results and therefore increase drug therapy safety....

[High activity antiretroviral therapy change associated to adverse drug reactions in a specialized center in Venezuela].
Subiela JD and Dapena E. Invest Clin 2016. PMID 27382797 Spanish.
Adverse drug reactions (ADRs) represent the first cause of change of the first-line highly active antiretroviral therapy (HAART) regimen, therefore, they constitute the main limiting factor in the long-term follow up of HIV patients in treatment. ...

Combining oral anticoagulation and antiplatelet therapies: appropriate patient selection
Vandiver JW and Diane Beavers K. J Thromb Thrombolysis 2018 - Review. PMID 29478128
Anticoagulant therapy and antiplatelet therapy are used regularly for prevention of arterial and venous thrombosis, and combinations of the two drug classes are seen with relative frequency in clinical practice. While co-prescribing is as high as 39-55% in some real-world cohort studies, the number of patients that meet criteria for combination therapy based on the overall body of evidence is likely much lower. ...
